Overview

Tumor-specific peptide-HLA complexes derived from Dark Antigens are antigenic complexes formed by the presentation of peptides, translated from previously silenced, noncoding, or "dark" regions of the genome, on the surface of cancer cells via HLA molecules. These peptides arise through epigenetic alterations or aberrant transcription associated with tumorigenesis, producing polypeptides that are rarely, if ever, expressed in normal tissues. Because they are shared across major solid tumor types, homogeneously expressed, and generally absent from healthy cells, these complexes offer attractive targets for immunotherapies—including engineered T cell therapies and cancer vaccines. Their unique presentation allows for potent and broad anti-tumor immune responses but necessitates careful selection to avoid off-target effects.
