Tumor suppressor candidate 1 (TUSC1) is a protein encoded by an intronless gene located on chromosome 9p21.2, identified as a putative tumor suppressor[1][2][3]. TUSC1 expression is often reduced or lost in cancer cells, including lung cancer and hepatocellular carcinoma, where downregulation is associated with tumor progression and adverse prognosis[1][2][3]. Restoration of TUSC1 expression in tumor cell lines suppresses cell proliferation in vitro and inhibits tumor growth in vivo, indicating its tumor suppressive capacity[1]. Loss of TUSC1 expression can result from mechanisms such as intragenic hypermethylation, making epigenetic status of the TUSC1 gene a promising prognostic biomarker in certain malignancies[2][3]. TUSC1 localizes to both the cytoplasm and nucleus, but its detailed biological pathways and interacting partners remain to be fully elucidated. Currently, no drugs are known to directly target TUSC1, nor is there evidence it serves as a direct therapeutic target (such as a receptor or enzyme).
