Overview

The tumor vasculature and tumor cell compartment represent two distinct but interdependent components of the tumor microenvironment targeted in oncology. The tumor vasculature consists of the network of blood vessels that supply oxygen and nutrients to the tumor, often characterized by leaky, disorganized structures driven by pro-angiogenic factors like VEGF (National Cancer Institute, 2023). The tumor cell compartment comprises the heterogeneous population of malignant cells that drive disease progression and metastasis. Therapeutic strategies targeting both compartments aim to disrupt the metabolic support system of the tumor while simultaneously exerting direct cytotoxic effects on the cancer cells (PubMed, PMID: 22430630). This dual approach is frequently utilized by multi-targeted tyrosine kinase inhibitors and certain ligand-directed toxins to overcome the limitations of monotherapy and reduce the likelihood of resistance (Journal of Clinical Oncology, 2021). By addressing both the "soil" (vasculature) and the "seed" (tumor cells), these therapies attempt to achieve more durable clinical responses in solid tumors.
