Overview

The tunica albuginea is a dense, fibrous envelope of connective tissue that encapsulates several anatomical structures, including the testes, ovaries, and the corpora cavernosa of the penis (StatPearls, 2023). In the male reproductive system, the tunica albuginea of the penis is a complex bilayered structure composed of collagen and elastin fibers that is essential for the veno-occlusive mechanism required to maintain an erection (PubMed, 2020). It is not a single molecular target or receptor, but rather an anatomical tissue layer that becomes the focus of therapeutic intervention in conditions like Peyronie's disease, where localized fibrosis and inelastic plaque formation occur (NIH, 2022). Drugs such as collagenase clostridium histolyticum are specifically indicated to treat these plaques by enzymatically breaking down the excessive Type I and Type III collagen that comprises the thickened tunica (FDA, 2013). Understanding the structural integrity and biomechanical properties of this tissue is crucial for managing penile trauma, erectile dysfunction, and fibrotic disorders of the reproductive organs.
