TUSC2 pseudogene 1 (TUSC2P1), also known as TUSC2P, is a non-coding pseudogene highly homologous to the 3′-UTR of the tumor suppressor candidate gene TUSC2 (also called FUS1). TUSC2P1 does not encode a protein, but instead functions by binding and sequestering microRNAs (miRNAs) that would otherwise suppress TUSC2 mRNA, thereby indirectly modulating TUSC2 protein levels. Expression of TUSC2P1 has been shown to inhibit cancer cell proliferation, migration, invasion, and colony formation, mainly through this miRNA decoy function. TUSC2P1 may also regulate other target genes sharing similar miRNA binding sites, such as TIMP2 and TIMP3. Reduced TUSC2P1 expression has been linked with increased malignancy in several cancer cell types. While TUSC2 (the canonical gene) is sometimes a therapeutic target or biomarker, TUSC2P1 is a regulatory pseudogene and not itself considered a direct therapeutic target or druggable protein [4].
