Overview

Tweety family member 2 (TTYH2) is a conserved, widely expressed membrane protein comprising five transmembrane domains. It was originally annotated as a calcium-activated large conductance chloride channel, but recent structural and functional studies indicate this may not be its main role: TTYH2 adopts a unique fold, lacks canonical ion channel pores, and forms distinct dimeric assemblies influenced by calcium, suggesting possible functions in protein complex formation or membrane interactions rather than direct anion conduction. TTYH2 is found on the plasma membrane, predominantly in the brain and certain cancers, and may be involved in cell signaling, cell adhesion, proliferation, and migration. Overexpression of TTYH2 is associated with poor prognosis in several cancers. There are currently no approved drugs targeting TTYH2, and its exact physiological function as a therapeutic target remains incompletely defined.
