Overview

TWIST1 associated long noncoding RNA regulated by androgen receptor (TANAR) is a specific long noncoding RNA (lncRNA) located on human chromosome 2 (240,981,515-240,986,072) whose expression is transcriptionally upregulated by the androgen receptor (AR)[2][5]. TANAR regulates the stability of TWIST1 mRNA by directly binding to its 5′ untranslated region (UTR), thereby competitively inhibiting the interaction of the nonsense-mediated mRNA decay (NMD) machinery component UPF1 with TWIST1 mRNA[2][3]. This leads to increased TWIST1 mRNA stability and protein expression. TANAR is implicated in cancer biology, particularly in clear cell renal cell carcinoma (ccRCC), where it enhances vasculogenic mimicry (VM), a tumor-associated neovascularization process that may contribute to tumor progression and metastasis[2][3][6]. Targeting the AR–TANAR–TWIST1 axis is being investigated as a strategy for anti-angiogenesis therapy in ccRCC. Currently, no therapeutic drugs are reported to directly target TANAR.
