Overview

Twisted gastrulation BMP signaling modulator 1 (TWSG1) is an evolutionarily conserved secreted glycoprotein that regulates the activity of bone morphogenetic proteins (BMPs), key growth factors in the transforming growth factor β (TGFβ) superfamily[2][1]. TWSG1 acts both as an antagonist and agonist of BMP signaling: it binds BMP ligands and the antagonist Chordin, forming complexes that modulate the availability and activity of BMPs in diverse developmental processes, including embryonic dorsoventral axis patterning, neural and craniofacial development, bone and cartilage formation, thymocyte development, and hematopoiesis[2][1][6][7]. Dysregulation of TWSG1 expression has been implicated in cancer (with roles varying depending on tissue context), bone diseases, immune system modulation, and fertility. TWSG1 is essential for proper development and tissue homeostasis; thus, interventions targeting TWSG1 must account for potential pleiotropic effects in various organ systems[2][1][5].

Mechanism of action

No approved drugs; however, theoretical mechanisms based on protein function: Inhibition of BMP signaling (by preventing BMP ligand-receptor binding through ternary complex formation with Chordin and BMPs); Activation of BMP signaling (by promoting Chordin cleavage and release of BMPs, or synergistic effects in certain contexts).
