Type 1 conventional dendritic cells (cDC1) are a specialized subset of dendritic cells that play a central role in orchestrating immune responses, particularly by promoting cytotoxic CD8+ T cell differentiation and mediating antitumor immunity. They are essential for initiating de novo T cell responses, cross-presenting exogenous antigens on MHC class I molecules, and supporting the activation of natural killer (NK) cells and NKT cells. Their abundance correlates with better patient survival rates in cancer and are required for effective rejection of immunogenic tumors and responsiveness to checkpoint blockade therapies.
Flt3 ligand promotes cDC1 development; checkpoint blockade therapies rely on cDC1 function to eliminate tumors; agonists targeting maturation pathways enhance cDC1 activation and function.
