Tc1 lymphocytes are a subset of CD8+ T cells that play a critical role in cell-mediated immunity, particularly against viral infections and tumors. They are characterized by their production of IFN-γ and TNF-α, as well as their potent cytolytic activity mediated by granzyme B and perforin. Differentiation into the Tc1 lineage is driven by IL-12 and IFN-γ, and regulated by transcription factors such as T-bet, Blimp-1 and IRF4. Imbalances in Tc1 populations can contribute to disease susceptibility, while adoptive transfer of Tc1 cells shows promise in cancer immunotherapy.
Tc1 cells exert their cytotoxic function primarily through the release of granzyme B and perforin, leading to target cell apoptosis. They also secrete IFN-γ and TNF-α, which contribute to immune regulation and direct anti-tumor effects.
