Type 1 Natural Killer T (NKT) cell activation refers to the process by which a specialized subset of T lymphocytes, known as type I or invariant NKT (iNKT) cells, are stimulated to exert their immunoregulatory functions. These cells bridge innate and adaptive immunity and are characterized by their semi-invariant T-cell receptor (TCR), which recognizes lipid antigens presented by the non-polymorphic MHC class I-like molecule CD1d. Activation can occur via TCR-mediated recognition of glycolipid antigens or cytokine-mediated mechanisms. Activated type 1 NKT cells release a variety of cytokines and chemokines, modulating immune responses in various contexts including antitumor immunity, regulation of chronic inflammation, and bridging innate/adaptive immunity. The balance between activating/inhibitory signals determines functional outcome, and overstimulation can lead to exhaustion. iNKT cell activation is a target for immunotherapy strategies in cancer and infectious diseases.
