Type 1 tumour necrosis factor receptor (TNFR1) is a transmembrane receptor that binds to tumor necrosis factor-alpha (TNFα) and regulates inflammatory pathways and immune responses. Upon binding with TNFα, TNFR1 activates intracellular signaling pathways involved in cell survival, apoptosis, and inflammation. It contains a death domain (DD) essential for recruiting adaptor proteins and initiating downstream signaling. Mutations in the TNFRSF1A gene are associated with tumor necrosis factor receptor-associated periodic syndrome (TRAPS). Elevated serum levels of TNFR1 have been linked to conditions such as schizophrenia, bipolar disorder, cognitive impairment, and dementia. TNFR1 plays a complex role in cancer, mediating both anti-tumor and pro-tumor effects. It is constitutively expressed on almost all nucleated cells.
