Overview

Type 2 DNA topoisomerase 6 subunit B-like (TOP6BL) is a component of the topoisomerase VI-like complex that is specifically required for the initiation of meiotic DNA double-strand breaks (DSBs), which are essential for homologous recombination, accurate chromosome pairing, and segregation during meiosis[1][2][3][4]. TOP6BL is the eukaryotic homolog of the archaeal Top6B subunit and forms a holocomplex with SPO11, enabling SPO11's endonucleolytic activity to generate DSBs in meiotic cells[2][3]. Loss of TOP6BL or its partner SPO11 results in meiotic arrest and infertility in mammals[2]. While TOP6BL is structurally related to classical type IIB topoisomerases, its physiological role is distinct and specialized for meiosis; it is not known to be a therapeutic or drug target[2][3][4]. No interacting drugs or clinical biomarkers are reported for TOP6BL at present[4]. Defects or loss of function in this complex are associated primarily with infertility due to failure of meiotic recombination rather than with cancer or other somatic diseases[2][3][4].
