Type 2 helper T cell activation refers to the process by which naïve CD4+ T cells are stimulated—primarily by antigen-presenting cells and specific cytokine environments—to differentiate into Th2 effector cells. This activation is typically associated with antigens that drive humoral (antibody-mediated) immune responses, including those from extracellular parasites and allergens. The process is orchestrated by engagement of the T-cell receptor (TCR) with antigen/MHC class II complexes, costimulatory signals (notably via CD28 and ICOS), and cytokine signaling—especially interleukin 4 (IL-4). Activated Th2 cells secrete a distinct cytokine profile (IL-4, IL-5, IL-9, IL-13, IL-25), which results in the activation of B cells (promoting IgE production), eosinophils, basophils, and mast cells, and plays key roles in allergic inflammation and protection against helminths. Overactivation or dysregulation of Th2 responses underlies conditions such as asthma and atopic dermatitis. Th2 activation is not a single protein or receptor but a complex, multi-step immune process.
