Overview

Type 2 immune response is a physiological immunological pathway specializing in defense against helminth parasites and environmental toxins, but it is also central to the pathology of many chronic allergic and inflammatory diseases such as asthma, atopic dermatitis, and chronic rhinosinusitis[1][2][3][4][6][7][8][9]. This response is characterized by the activation of type 2 helper T cells (Th2), innate lymphoid cells (ILC2), and the production of cytokines such as interleukin-4, -5, and -13, which drive IgE class switching in B cells, promote eosinophil recruitment, and induce mucosal barrier and tissue changes[1][3][6][7][8][9]. Numerous therapeutics (biologics) have been developed to target components of type 2 immunity for diseases with underlying type 2 inflammation, but the response itself is a complex immunological process, not a discrete molecule, receptor, or protein therapeutic target. Thus, "Type 2 immune response" does not refer to a single molecular entity or direct drug target, but rather to an orchestrated immune pathway encompassing multiple cell types, signaling molecules, and effector mechanisms[1][2][4][6][7].
