Overview

Type 2 phosphatidylinositol-4,5-bisphosphate 4-phosphatase (PIP4P2) is a transmembrane enzyme localized to late endosome and lysosome membranes, where it catalyzes the removal of the 4-phosphate group from phosphatidylinositol-4,5-bisphosphate (PtdIns-4,5-P2), producing phosphatidylinositol-4-phosphate (PtdIns-4-P)[2][5][6]. It is specific for PI(4,5)P2, does not act on other phosphoinositide forms such as PI(3,4,5)P3 or PI(3,4)P2[2][5]. PIP4P2 negatively regulates phagocytosis and impacts cellular signaling by controlling levels of PI(4,5)P2, which is central to numerous processes, including cytoskeletal dynamics, membrane trafficking, and ion channel regulation[2][4][6]. Genetic variations are associated with inherited neurologic disorders such as spinocerebellar ataxia[2].
