Type I nitroreductases are oxygen-insensitive flavoenzymes that catalyze the two-electron reduction of nitroaromatic compounds and quinones, utilizing NADH or NADPH as electron donors. They are found in bacteria and some eukaryotes, including protozoan parasites. They play a role in xenobiotic detoxification, prodrug activation (e.g., in antiparasitic therapy), and are exploited in gene-directed enzyme prodrug therapy (GDEPT) for cancer treatment. Unlike Type II nitroreductases, they do not generate reactive oxygen species.
