Type II bacterial DNA topoisomerases are essential enzymes that manage the topology of DNA in prokaryotic cells. They play a critical role in processes such as DNA replication, transcription, recombination, and chromosome segregation by introducing transient double-stranded breaks into the DNA helix and passing another segment of duplex DNA through these breaks. They are validated targets for antibacterial drugs.
Inhibition of DNA gyrase or topoisomerase IV by targeting the catalytic core or ATP-binding site, leading to DNA double-strand breaks and cell death.
