Overview

Type II bacterial topoisomerases are essential enzymes that manage the topology of DNA in prokaryotic cells. They play a critical role in processes such as DNA replication, transcription, recombination, and chromosome segregation by introducing transient double-stranded breaks into the DNA helix and passing another segment of duplex DNA through these breaks before resealing them. This mechanism allows them to resolve supercoils, knots, and catenanes that arise during cellular processes. DNA gyrase (a Type IIA topoisomerase) is unique because it can introduce negative supercoils into relaxed circular DNA. Because their function is vital for bacterial cell viability, these enzymes are prime targets for antibacterial drugs.
