Overview

Thyroxine 5'-deiodinase (DIO3) is an enzyme that inactivates thyroid hormones by catalyzing the removal of an iodine atom from the inner ring of T4 and T3. It is highly expressed in the placenta, fetal tissues, and neonatal tissues, playing a critical role in regulating thyroid hormone levels during development. By converting T4 to rT3 and T3 to T2, DIO3 prevents premature exposure of developing tissues to excessive thyroid hormone concentrations.
