Tyrosinase-derived peptide–HLA class II complexes are specialized molecular structures formed when fragments of the tyrosinase enzyme, a key protein in melanin biosynthesis, are processed and presented on the surface of cells by Human Leukocyte Antigen (HLA) class II molecules. While tyrosinase is primarily expressed in melanocytes, it is significantly overexpressed in melanoma cells, making these complexes vital targets for cancer immunotherapy. Unlike class I complexes that interact with CD8+ cytotoxic T cells, these class II complexes are recognized by CD4+ helper T cells, which play a critical role in coordinating a robust and sustained anti-tumor immune response. Therapeutic strategies targeting these complexes include peptide-based vaccines and adoptive cell therapies using T-cell receptors (TCRs) engineered to recognize specific tyrosinase epitopes, such as those presented by HLA-DRB1*04:01. However, because tyrosinase is also present in normal melanocytes, targeting these complexes can lead to autoimmune side effects like vitiligo or ocular inflammation.
