Tyrosinase is a copper-containing oxidase and the rate-limiting enzyme of melanin synthesis, catalyzing conversion of tyrosine to DOPA and DOPA to dopaquinone as critical steps in pigment formation. The TYR gene encodes this enzyme, and mutations cause oculocutaneous albinism type 1 in humans. Tyrosinase mRNA is the transcript measured to estimate enzyme expression levels in research and clinical diagnostics, particularly in melanoma, where elevated mRNA/protein levels are associated with disease progression and prognosis. While tyrosinase mRNA is targeted in siRNA-based research, the direct therapeutic target in clinical settings is the enzyme or its gene. Inhibitors of tyrosinase are used therapeutically or cosmetically to reduce melanin production in hyperpigmentation disorders.
