Overview

Tyrosinase-related protein 1 (TRP-1), also known as gp75, is a 75 kDa transmembrane glycoprotein primarily localized to the melanosomes of melanocytes and melanoma cells (UniProt P17642). It serves a critical role in the melanin biosynthetic pathway by acting as a 5,6-dihydroxyindole-2-carboxylic acid (DHICA) oxidase, which facilitates the conversion of DHICA to indole-5,6-quinone-2-carboxylic acid (PubMed: 15373230). Beyond its enzymatic activity, TRP-1 is vital for the stabilization of tyrosinase and the maintenance of melanosome integrity. In oncology, TRP-1 is recognized as a tumor lineage antigen because it is highly expressed in the majority of melanomas while remaining restricted to pigmented cells (PubMed: 22415316). This specificity makes it a target for immunotherapeutic interventions, including monoclonal antibodies like IMC-20D7S and various cancer vaccines designed to elicit a robust anti-tumor immune response (PubMed: 25214086). A primary clinical challenge associated with targeting TRP-1 is the development of vitiligo, an autoimmune condition resulting from the destruction of healthy melanocytes (PubMed: 18483247).

Monoclonal antibodies targeting TRP-1 induce antibody-dependent cellular cytotoxicity (ADCC) against melanoma cells, while vaccines aim to stimulate T-cell mediated immune responses against the antigen (PubMed: 22415316, 25214086).
