Overview

The Tyrosinase-related protein 1-specific T-cell receptor (TRP-1 TCR) is an immune receptor found on CD8+ T cells that recognizes TRP-1-derived peptides presented by MHC class I molecules. TRP-1, also known as gp75, is a glycoprotein essential for melanin synthesis and is a prominent melanoma-associated antigen (PubMed: 15155838). While TRP-1 is a well-known target for CD4+ T cells, specific MHC class I-restricted epitopes have been identified, allowing for the development of TRP-1-specific CD8+ TCR-T cell therapies (PubMed: 21602451). These engineered T cells are designed to recognize and eliminate melanoma cells by inducing apoptosis through the release of cytotoxic granules (PubMed: 28923845). Clinical application of this target is primarily focused on metastatic melanoma, though it faces challenges regarding 'on-target, off-tumor' toxicity against healthy melanocytes in the skin and eyes (PubMed: 19075010). Monitoring for TRP-1 expression and specific HLA alleles is crucial for patient selection in these immunotherapeutic approaches.

Mechanism of action

The TCR specifically binds to TRP-1 peptide fragments presented by MHC class I molecules, initiating a signaling cascade through the CD3 complex that results in T-cell proliferation, cytokine production, and the directed lysis of TRP-1-expressing cells.
