Overview

Tyrosinase-related protein 2 (TRP-2) peptide presented by H2-Kb is a specific peptide-major histocompatibility complex (pMHC) that serves as a prominent tumor-associated antigen in murine melanoma models (Bloom et al., J Exp Med, 1997). TRP-2, also known as dopachrome tautomerase (DCT), is an enzyme essential for melanin biosynthesis and is expressed in both healthy melanocytes and malignant melanoma cells (Parkhurst et al., Cancer Res, 1998). The specific epitope TRP-2(180-188), characterized by the amino acid sequence SVYDFFVWL, is processed and presented on the cell surface by the H2-Kb class I molecule. This complex is recognized by the T-cell receptor (TCR) of specific CD8+ cytotoxic T lymphocytes, making it a primary target for immunotherapeutic interventions such as peptide vaccines, DNA-based vaccines, and adoptive cell transfer (Schreurs et al., Cancer Res, 2000). Because TRP-2 is a self-antigen, therapeutic targeting must overcome immunological tolerance, which often results in autoimmune-mediated destruction of normal melanocytes, manifesting as vitiligo (Overwijk et al., J Exp Med, 1999). The TRP-2/H2-Kb interaction is widely utilized in preclinical oncology to study the mechanisms of T-cell activation, tumor escape, and the efficacy of novel immunotherapy platforms in C57BL/6 mice.
