Overview

Tie2, also known as TEK tyrosine kinase, is a receptor tyrosine kinase predominantly expressed in endothelial cells. It plays a critical role in the development, remodeling, and maintenance of blood and lymphatic vessels. Tie2 is essential for angiogenesis and vascular homeostasis. Its activity is tightly regulated by its ligands, the angiopoietins. Upon activation, Tie2 triggers several key pathways involved in cell survival, migration, proliferation, and vessel stability. Dysregulation or mutation of Tie2 has been implicated in pathological conditions such as tumor growth/metastasis, congenital glaucoma, lymphatic malformations, inflammation, and other vascular disorders.
