Tyrosine-protein kinase non-receptor type 2 (TYK2) is a member of the Janus kinase (JAK) family and functions as a non-receptor tyrosine kinase. It plays an essential role in signal transduction for various cytokines and interferons, regulating processes such as cell growth, development, migration, and immune responses. TYK2 mediates signaling downstream of several cytokine receptors including those for IFN-alpha/beta (type I interferons), IL-6, IL-10, IL-12, IL-23, among others. Deficiency or dysfunction of TYK2 can lead to significant immunological consequences, while aberrant activation or mutation may contribute to inflammatory disorders or malignancies. TYK2 has become an important therapeutic target for drug development aimed at treating autoimmune diseases such as rheumatoid arthritis or psoriasis by modulating JAK–STAT pathway activity.
TYK2 becomes activated upon association with specific cytokine receptors. Upon ligand binding to these receptors: - The receptor-associated TYK2 undergoes autophosphorylation. - This leads to phosphorylation of downstream targets such as STAT proteins. - The activated STATs translocate to the nucleus to regulate gene expression involved in immune modulation and inflammation
