RET (Rearranged during Transfection) is a receptor tyrosine kinase that plays a crucial role in cell signaling, particularly in the development of the nervous system and kidneys, as well as in spermatogenesis. It is activated by glial cell line-derived neurotrophic factor (GDNF) family ligands that bind to GFRα coreceptors, forming a ternary complex essential for downstream signaling. Mutations or alterations can lead to loss-of-function mutations causing Hirschsprung’s disease or gain-of-function mutations causing oncogenic activation in several cancers.
