Overview

Tyrosine-protein kinase receptor Tie2 is a receptor tyrosine kinase expressed almost exclusively in endothelial cells. It plays crucial roles in angiogenesis, lymphangiogenesis, and vascular development. Activation occurs through binding to angiopoietin ligands, leading to dimerization and kinase activation. Defects in Tie2 are associated with inherited venous malformations and primary congenital glaucoma. In cancer, Tie2 is expressed in tumor-associated monocytes and contributes to tumor angiogenesis.

Mechanism of action

Ligand binding induces dimerization and trans-phosphorylation of tyrosine residues, leading to activation of the kinase domain and downstream signaling.
