Tyrosine-protein kinase SYK is a non-receptor type protein-tyrosine kinase containing two Src homology 2 (SH2) domains in tandem and multiple autophosphorylation sites. It is activated upon binding of its SH2 domains to phosphorylated ITAMs and plays essential roles in lymphocyte development, immune cell activation, cell adhesion, and vascular development. SYK regulates several biological processes by mediating tyrosine phosphorylation of multiple proteins involved in calcium mobilization and MAPK cascades. It is a key molecule controlling multiple physiological functions and has been implicated in the pathogenesis of malignant cancer.
