Overview

Tyrosine-protein kinase TEK (TEK, also known as TIE2) is a receptor tyrosine kinase primarily expressed in endothelial cells lining blood vessels. It plays a crucial role in mediating communication between endothelial cells and smooth muscle cells, directing blood vessel formation (angiogenesis), and ensuring the structural integrity of blood vessels. Upon activation by its main ligand angiopoietin-1 (ANGPT1), the receptor triggers intracellular signaling cascades that regulate endothelial cell survival, proliferation, migration, and differentiation. Mutations in *TEK* are linked to multiple cutaneous and mucosal venous malformations (VMCM). In cancer biology, Tie2 expressing monocytes can promote tumor angiogenesis.
