Overview

TIE2 is a receptor tyrosine kinase expressed primarily in vascular endothelial cells and some monocytes. It plays a crucial role in angiogenesis, vascular development, and maintaining vascular stability by interacting with angiopoietins. Activation of TIE2 promotes endothelial cell survival, proliferation, and migration through the PI3K-Akt pathway. Mutations in TIE2 are associated with venous malformations and it contributes to tumor neovascularization.

Mechanism of action

Ligand binding (Ang1, Ang2, Ang4) induces TIE2 dimerization and activation of downstream signaling pathways, including PI3K-Akt.
