Overview

Tyrosine-protein phosphatase non-receptor type 18 (PTPN18) is an intracellular enzyme of the protein tyrosine phosphatase family, encoded by the PTPN18 gene. It acts by dephosphorylating tyrosine residues on target proteins, which regulates key cellular processes including growth, differentiation, cell cycle, and signal transduction. PTPN18 contains a PEST motif implicated in protein-protein interactions and intracellular stability. Its subcellular localization (nuclear vs. cytoplasmic) appears to determine its function as a tumor suppressor (notably in breast and hepatocellular cancer) or as a potential promoter of malignancy (notably in colorectal and endometrial cancer). It has been shown to regulate autophosphorylated tyrosine kinases such as HER2, but its detailed substrates and functions are still under investigation. Currently, there are no specific approved drugs or established clinical biomarkers associated directly with PTPN18
