Overview

Ubiquitin-associated domain-containing protein 1 (UBAC1) is the non-catalytic subunit of the Kip1 ubiquitination-promoting complex (KPC), an E3 ubiquitin-protein ligase complex. The KPC complex mediates polyubiquitination and subsequent proteasomal degradation of proteins such as CDKN1B—a cyclin-dependent kinase inhibitor critical for cell cycle control during the G1 phase. UBAC1 functions as an adapter, facilitating the transfer of polyubiquitinated substrates to the 26S proteasome for degradation. It also regulates the NF-kappa-B signaling pathway by promoting maturation of NFKB1. UBAC1 is localized to the cytosol, Golgi apparatus, and plasma membrane. While it is associated with protein modification, maturation, and degradation processes, it is not a direct therapeutic drug target, and there are no FDA-approved drugs, clinical biomarkers, or reported safety concerns linked specifically to UBAC1[1][9][4][8].
