Ubiquitin B pseudogene 4 encodes several ubiquitin protein variants that differ in amino acid sequence from canonical ubiquitin, most notably the variant called Ub KEKS (Q2K, K33E, Q49K, N60S)[2]. Unlike canonical ubiquitin, which marks proteins for proteasomal degradation, Ub KEKS modifies targets such as nuclear lamins, with these conjugates not being targeted for degradation[2]. Knockout of UBBP4 in human cell lines delays cell division and causes accumulation of lamin A in the nucleolus, indicating roles in cell cycle regulation and nuclear architecture[2]. Originally annotated as an unprocessed pseudogene, UBBP4 has now been shown through transcriptomic and proteomic analyses to produce functional proteins in humans[2][3][5][7]. The presence of ‘pseudogene’ in its name can be misleading, as it encodes proteins that participate in cellular processes, but there is currently no evidence that it serves as a direct therapeutic drug target.
