Overview

Ubiquitin C-terminal hydrolase L1 (UCHL1) mRNA is the transcript responsible for the synthesis of the UCHL1 protein, a highly abundant neuronal enzyme involved in the ubiquitin-proteasome system (UPS). UCHL1 functions primarily as a deubiquitinating enzyme, recycling ubiquitin monomers to maintain cellular proteostasis, and is essential for neuronal health (UniProt P09936). Dysregulation of UCHL1 mRNA levels is associated with several neurodegenerative conditions, most notably Parkinson's disease, where decreased expression is linked to protein aggregation and neuronal death (PubMed: 12446708). In the context of drug development, UCHL1 mRNA is a target for novel RNA-based therapeutics such as SINEUPs, which are long non-coding RNAs designed to bind the mRNA and selectively enhance its translation to restore protein levels (Nature: 10.1038/nature11546). Conversely, in certain cancers where UCHL1 is overexpressed and promotes tumor progression, the mRNA may be targeted for degradation using antisense oligonucleotides or siRNA (PubMed: 28651074). Monitoring UCHL1 levels in biofluids also serves as a critical biomarker for traumatic brain injury and neurodegeneration (NIH: 10.1016/j.clinbiochem.2016.01.018).
