Ubiquitin Carboxyl-Terminal Hydrolase L5 (UCHL5) is a cysteine protease and deubiquitinating enzyme (DUB) that cleaves 'Lys48'-linked polyubiquitin chains from substrate proteins. It is associated with the 19S regulatory subunit of the 26S proteasome and plays a key role in protein degradation. UCHL5 participates in various cellular processes, including protein deubiquitination, regulation of primary metabolic processes, and positive regulation of the smoothened signaling pathway. Overexpression has been linked to various cancers, particularly endometrial cancer, through Wnt/β-catenin pathway activation. Dysregulation may also play roles in neurodegeneration.
