Overview

Ubiquitin-conjugating enzyme E2 S (UBE2S) is a member of the E2 ubiquitin-conjugating enzyme family, catalyzing the attachment of ubiquitin to substrate proteins. UBE2S is a key component of the anaphase-promoting complex/cyclosome (APC/C), controlling cell cycle progression through mitosis by elongating Lys-11-linked polyubiquitin chains on specific substrates, thereby targeting them for proteasomal degradation and promoting mitotic exit. It also modulates the stability of important signaling proteins such as β-catenin, enhancing their accumulation via K11-linked polyubiquitination and activating the Wnt/β-catenin pathway. UBE2S is overexpressed and functionally implicated in various cancers, where it contributes to malignant cell proliferation, migration, and poor prognosis. Inhibitors of related pathways, such as ETC-159, may indirectly target UBE2S function[1][3][5][7][8].
