Ubiquitin conjugating enzyme E2 T (UBE2T)

Target
UBE2T
Molecular classification
Enzyme, Ubiquitin-conjugating enzyme (E2), Member of ubiquitin-proteasome system
01

Overview

Ubiquitin conjugating enzyme E2 T (UBE2T) is an E2 class enzyme central to the post-translational modification of proteins via ubiquitination, serving a pivotal role in the Fanconi anemia DNA repair pathway through its association with E3 ligase FANCL and its ability to monoubiquitinate critical substrates such as FANCD2 and FANCI. Deficiency or mutation of UBE2T leads to Fanconi anemia (subtype FA-T), and its dysregulation is linked to cancer development, metastatic behavior, cell cycle control, apoptosis, and resistance to therapy. Drugs and inhibitors targeting UBE2T are under preclinical study, highlighting its value as a molecular target in oncology and hematologic disorders[1][2][4][5][6].

Mechanism of action

Small molecule inhibition: blocks UBE2T-mediated degradation of RACK1, suppresses Wnt/β-catenin signaling, inhibits proliferation and invasion siRNA and genetic knockdown: G2/M cell cycle arrest, increased apoptosis

04

Disease associations

Cancer (oncogenesis, metastasis, therapy resistance)Fanconi anemia (FA, specifically FA complementation group T)Potential roles in other disease processes through dysregulation of DNA repair, cell cycle, and apoptosis
05

Safety considerations

Targeting UBE2T could impact DNA repair fidelity and genomic stability, leading to potential cytotoxicity or secondary malignanciesRisk of affecting normal cell cycle and apoptosis
