Overview

Ubiquitin-conjugating enzyme E2 T (UBE2T) is a critical E2 enzyme within the Fanconi anemia (FA) DNA repair pathway, where it facilitates the monoubiquitination of the FANCD2-FANCI complex (UniProt Q9NPD8). This enzymatic step is essential for the repair of DNA interstrand cross-links, thereby ensuring genomic stability during replication (NCBI Gene: 29089). UBE2T is frequently overexpressed in a wide range of malignancies, including breast, lung, and hepatocellular carcinomas, where it acts as an oncogene to promote cell proliferation, epithelial-mesenchymal transition, and resistance to DNA-damaging chemotherapy (PubMed: 30603351). Consequently, UBE2T mRNA has emerged as a significant therapeutic target, with strategies such as small interfering RNAs (siRNAs) and antisense oligonucleotides (ASOs) being developed to downregulate its expression (PubMed: 32853215). Additionally, small molecule inhibitors are being explored to block the protein's catalytic activity and sensitize cancer cells to treatment.
