Overview

Ubiquitin domain-containing protein 1 (UBTD1) is an evolutionarily conserved protein with a ubiquitin-like domain that acts as a scaffold in the ubiquitin-proteasome system, enabling or enhancing the interactions between E2 conjugating enzymes and E3 ligases, thereby modulating protein ubiquitination and degradation[1][4][5][6]. UBTD1 is cytoplasmic and membrane-associated, localizing to cell–cell adhesion sites, and plays essential roles in cellular signal transduction by regulating the stability of key proteins such as yes-associated protein (YAP), p53, MDM2, ASAH1, and others[1][3][6]. It controls major regulatory points in signaling pathways including EGFR phosphorylation and degradation, YAP/TAZ activity, and actomyosin contractility pathways like RhoA/ROCK[1][3][4]. UBTD1 depletion exacerbates EGFR signaling, encourages cell proliferation, and increases cancer cell invasiveness, indicating a tumor suppressor role[1][3][6]. Its expression is downregulated in several human cancers and low expression correlates with increased cancer aggressiveness and reduced patient survival. Despite its prominent cellular functions, UBTD1 is not currently a direct therapeutic target, though its regulatory influence on cancer-related signaling pathways may confer future translational relevance[1][3][6].
