UBX domain-containing protein 10 (UBXN10)

UBX domain-containing protein 10 (UBXN10) is a member of the UBX-domain protein family, which displays structural similarity to ubiquitin. The UBX domain, consisting of approximately 80 amino acids, is found in a variety of eukaryotic proteins that function as cofactors or regulators in ubiquitin-related processes but is not conjugated to proteins in the manner of ubiquitin[2][4]. UBXN10 specifically acts as a tethering factor for VCP/p97, a AAA+ ATPase involved in diverse cellular activities, including protein degradation and organelle biogenesis. UBXN10 is essential for the assembly of cilia, localizing to both cilia and the intraciliary transport particle B complex, and helping recruit VCP/p97 to support ciliary functions[1][3][7]. Diseases associated with UBXN10 mutations include inclusion body myopathy with early-onset Paget disease of bone and spastic paraplegia. There is currently no evidence that UBXN10 serves as a direct therapeutic target or interacts with approved pharmacological agents. UBX domain-containing proteins help specify substrate specificity for VCP/p97, and may influence the ubiquitin-proteasome pathway and related cellular processes, though their function is distinct from canonical ubiquitin conjugation[1][2][3][4][7].
