Overview

UBX domain protein 6 (UBXN6) is a cytosolic adaptor protein of the UBX-domain family that modulates diverse cellular pathways, principally by linking the AAA-ATPase p97 (also called VCP) to specific ubiquitinated protein complexes at cellular membranes[1][3]. It plays a pivotal role in the ubiquitin–proteasome system by directing p97 to mono-ubiquitinated cargo such as caveolin-1, promoting their endosomal trafficking and degradation, and regulating ER-associated degradation (ERAD) of misfolded proteins[1][3]. UBXN6 also coordinates the removal of ubiquitin conjugates during autophagosome formation, contributing to clearance of damaged organelles and maintaining cellular proteostasis[1][3]. Functionally, UBXN6 acts as a positive regulator of JAK-STAT1/2 signaling by binding to the kinase TYK2 and stabilizing both TYK2 and the type I interferon receptor IFNAR1, thus promoting efficient immune signaling and antiviral responses[2]. Genetic alterations or dysregulation of UBXN6 are implicated in neuromuscular and neurodegenerative disorders, as well as immune defects and potentially certain cancers[2][3]. UBXN6 is not a typical direct drug target (receptor, enzyme, etc.), but may become relevant in therapeutic strategies aimed at modulating protein homeostasis, autophagy, or immune responses[1][2][3].
