UBX domain protein 8 (UBXN8) is a transmembrane protein localized to the endoplasmic reticulum (ER) membrane, where it acts as a cofactor for p97/VCP (valosin-containing protein). Through its UBX domain, it tethers p97 to the ER membrane, facilitating efficient ER-associated degradation (ERAD) of misfolded lumenal proteins—an important process in maintaining protein homeostasis. UBXN8 also displays ubiquitin-binding activity and may have a role in reproduction, although this function is less well characterized. While genetic variants have been linked to hereditary spastic paraplegia (SPG8), UBXN8 is not widely accepted as a direct drug target or biomarker.
