Overview

LpxC (UDP-3-O-(R-3-hydroxymyristoyl)-N-acetylglucosamine deacetylase) is an essential zinc-dependent metalloamidase enzyme found in virtually all Gram-negative bacteria. It catalyzes the first committed step in the biosynthesis of lipid A, a critical component of lipopolysaccharides (LPS). Lipid A acts as an endotoxin and is vital for bacterial viability and pathogenicity. As it catalyzes an essential step unique to bacteria, LpxC is a target for novel antibiotics. Inhibitors disrupt bacterial growth by blocking lipid A synthesis.
