Overview

UDP-GlcNAc--undecaprenyl-phosphate GlcNAc-1-phosphate transferase, commonly referred to as WecA or TagO, is an essential integral membrane enzyme found in a wide range of bacteria [1, 2]. It catalyzes the first committed step in the biosynthesis of various cell envelope glycoconjugates, including the O-antigen of lipopolysaccharides, the enterobacterial common antigen, and wall teichoic acids [1, 3]. The enzyme facilitates the transfer of N-acetylglucosamine-1-phosphate from UDP-GlcNAc to the lipid carrier undecaprenyl phosphate, creating a lipid-linked precursor necessary for further glycan assembly [2]. Because these glycoconjugates are vital for bacterial structural integrity, pathogenesis, and survival under environmental stress, this enzyme is a significant target for the development of novel antimicrobial agents [3]. However, a major challenge in targeting this enzyme is the existence of a functional human homolog, DPAGT1, which is involved in N-linked glycosylation; inhibitors like tunicamycin often lack the necessary selectivity, leading to host toxicity [4]. Consequently, current drug discovery efforts are focused on identifying highly selective inhibitors that can bypass the human ortholog to treat resistant bacterial infections [3, 4].
