UDP-N-acetylglucosamine enolpyruvyl transferase, commonly known as MurA, is a key bacterial enzyme that catalyzes the first committed step in peptidoglycan biosynthesis. Peptidoglycan is an essential component of the bacterial cell wall, making MurA a critical target for antibacterial drug development. Inhibition of MurA disrupts cell wall synthesis leading to bacterial death, it remains an attractive target for novel antibacterial agents.
Fosfomycin acts as a covalent inhibitor of MurA by modifying an active site cysteine residue, thereby blocking its enzymatic activity.
