UDP-N-acetylmuramoyl-tripeptide--D-alanyl-D-alanine ligase (MurF) is an essential bacterial enzyme involved in the biosynthesis of peptidoglycan, a critical component of the bacterial cell wall. It catalyzes the final step in the formation of UDP-N-acetylmuramoyl-pentapeptide, which serves as a precursor for murein (peptidoglycan) synthesis. MurF is responsible for catalyzing the ATP-dependent addition of D-Ala–D-Ala dipeptide to UDP-N-acetylmuramoyl-tripeptide during peptidoglycan biosynthesis. The activity of MurF is crucial for maintaining cell wall integrity and thus bacterial viability. It is an attractive target for antibacterial drug development due to its essential role and lack of human homologs.
