Uncharacterized cell-surface receptors refer to a broad and heterogeneous group of membrane-bound proteins that possess the structural characteristics of receptors but lack identified endogenous ligands or clearly defined biological roles (Alexander et al., 2023). These proteins, which include a significant number of orphan G protein-coupled receptors (GPCRs), are often identified through high-throughput genomic sequencing and are considered part of the "dark" druggable genome (Insel et al., 2019). Because they are situated on the cell surface, they are theoretically accessible to therapeutic agents and represent a vast frontier for novel drug discovery in areas such as oncology, metabolic diseases, and neuropharmacology (Hauser et al., 2018). However, the lack of functional characterization makes it impossible to predict the physiological consequences of modulating these proteins without further research. Current efforts in the pharmaceutical industry focus on "deorphanization"—the process of pairing these receptors with their natural signaling molecules—to unlock their therapeutic potential. As a collective term, it does not describe a single molecular entity but rather a category of proteins awaiting functional validation.
