KIAA1671 is a large (∼196 kDa) and largely disordered protein found in humans, initially identified in large-scale genomic and proteomic studies. Recent spatial proteomics and cell imaging work reveal that it is a resident protein of the brush border actin cytoskeleton in polarised intestinal epithelial cells: it localizes specifically to the subapical terminal web at the base of microvilli, and targets F-actin–rich networks in both polarized and nonpolarized cell types. The protein is implicated in the initiation and formation of microvilli, acting in close association with EPS8 and potentially other assembly factors during early stages of microvillar growth. It associates dynamically with F-actin-rich puncta, suggesting a possible role in actin polymerization, though it lacks canonical actin-binding motifs. Bioinformatics and proteomic data indicate potential interaction with capping protein (CAPZA2) and possible regulation of filament growth. KIAA1671 remains largely uncharacterized outside these recent cellular studies and is not considered a classical drug target, receptor, or enzyme to date[2][3].
